Hydrophobic Interaction Chromatography (GFP Purification part 4 of 6)

When all of the eluate has been collected and pooled, it's time for HIC. Hydrophobic Interaction Chromatography is based on the principle that hydrophobic chemicals on the resin surface will bind to hydrophobic patches on the GFP protein. In order for this to occur, the resin and protein eluate have to be in a high salt environment to remove the water shielding. The salt we use is ammonium sulfate. To remove the attached GFP protein from the HIC column we lower the salt concentration during elution causing the water shielding to reform and the GFP protein detaches from the resin into the elution stream. The protein-rich eluate is collected and pooled. The product is now ready for the last major step, TFF.

For over 50 years, immunoassays have been used by physicians, biologists, and forensic scientists to diagnose disease, detect contamination, and to determine the origin of biological samples found at crime scenes. Immunoassays rely on the uniqueness of antibodies. Antibodies are typically produced by the immune system in response to antigens – bacteria, viruses, pollen, or anything that your body perceives as foreign. Our bodies are capable of producing billions of different protective proteins called antibodies, each one specific to a different antigen. When an antibody and an antigen are a match, they bind together – similar to a lock and key. Scientists are able to produce and purify antibodies – often from animals – that then can be used to test for many different things. The purpose of this activity is to develop student interest, knowledge, and skills in the areas of biotechnology and life sciences. It engages students with a hands-on activity that is fun, interesting, and informative. Join us as we explore a scientific explanation to what's lurking in the lake.

The cells, once again in paste form, are ready for the next step - Cell Disruption, also called Lysing. The cells are resuspended in a buffered solution and then pumped at high pressure, 900 bar - which is about 13,000 pounds per square inch - through the Homogenizer. Inside the Homogenizer they're forced through a tiny orifice. Just like a balloon being tightly squeezed, the cells can't take the stress - and they rupture and break apart. And to ensure that all the E.coli cells are ruptured, the solution is cycled through the homogenizer a second time.

All cells require nutrition. A properly formulated media contains the necessary nutrients to allow cells to grow and produce. Yeast Extract Tryptic Soy Broth Ammonium chloride Sodium bi-phosphate Monopotassium phosphate and an Antifoam compound Once all of these ingredients are in, another 10 kilograms of High Purity Water is added...all ports and valves are closed...all condensate valves are opened...and the bioreactor begins an SIP – Sterilize-In-Place cycle. The target for sterilization is 121 degrees Celsius for 30 minutes. As soon as that target is reached, the condensate valves are closed, and the SIP cycle completes automatically. Both the vessel and the media are now sterile…And we're ready to add the final ingredients to our media. The glucose hose is attached to the vessel. The connection is steamed to sterilize it, and the separately sterilized glucose-antibiotic solution is pumped into the vessel. Then a manual pH reading of the media is taken, and the bioreactor is set up for its fermentation

In order to grow, cells need very specific environmental conditions such as food, energy, proper temperature and humidity. When growing cells in the lab, we have to create these conditions using culture media. Solid culture media is a mixture of agar and nutrients poured into Petri dishes. We can transfer cells to the Petri dishes where they will grow with proper incubation. Sterile technique is required to create culture media so you don't have unwanted microorganisms growing in your culture. In this video, we'll demonstrate the steps for preparing Brain Heart Infusion Agar. It's a general-purpose media used for a variety of bacterial and fungal species including pathogens like Streptococcus and Pneumococcus.
