This animation demonstrates how DNA microarray experiments are performed. One common use of microarrays is to determine which genes are activated and which are repressed when two populations of cells are compared. Every gene is measured simultaneously. As an example, we'll compare what happens to yeast genes when cells are grown in aerobic versus anaerobic conditions.

EASE (Effective Authentic Science Event) is a one-day outreach program for middle school students with the goal of showing them that science is fun, and that the local community college is a good first choice for STEM education. Elution (GFP Purification part 3 of 6)

When all the lysate has entered the column – or when the capacity of the beads to bind the protein has been reached, it's time for Elution. Elution is the release of GFP from the beads with a new solution – in this case, a buffer that includes sodium-chloride solution. As the new buffer is pumped through the beads, at some point the GFP no longer binds to the beads and is released into the buffer. The resulting product stream is usually referred to as the Eluate. The UV optical density sensor measures protein concentration and indicates when product begins eluting from the column. At this point, outlet valves are switched to allow flow of the eluate to a collection vessel. When the UV sensor indicates when all of the GFP has come off of the chromatography resin, the outlet valves are switched to waste.

The Fermentation Cycle (GFP Fermentation part 6 of 6)

After the inoculation hose is connected to the reactor and steamed for 20 minutes, the expanded seed stock is pumped into the reactor containing the media. And now fermentation begins. The operator takes zero hour readings and monitors batch temperature, agitator RPMs, dissolved oxygen levels, pH, vessel pressure, optical density, air flow rate and glucose concentrations. When the targeted levels of glucose and optical density are achieved, it's time to add IPTG to the vessel to "turn on" the expression of the Green Fluorescent Protein in the cells. After about 5 hours, final readings are taken and a sample is drawn to check the percentage of cell solids. We now refer to the product as broth…it contains spent media and cells. The process is complete when the glucose is mostly consumed, and the batch has reached the desired concentration. The batch is then cooled down, pumped into a broth tank...and labeled with the batch number, volume, time and date. The harvested broth will now move downstream to Recovery where the cells will be ruptured to free the GFP…And the protein will be separated from the other broth components.

Fermentation Prep (GFP Fermentation part 4 of 6)

Before the fermentation process can begin, the area has to be prepared. All unneeded materials are removed. Equipment is cleaned and sterilized according to the standard operating procedures. All required materials and documentation should be gathered and prepared. And all Process Control software loaded and verified. The Fermentation batch process will be guided and documented with the BPR – Batch Process Record. The Batch Record leads the operator through the process, with each step requiring a sign-off and separate verification. This record also includes spaces for documenting key times, activities and instrument readings. Operators begin with a complete check of all critical equipment. Valves, caps and lines are checked, hoses are tightened, probes are verified and calibrated, and 10 kilograms of HPW – High Purity Water - is added to the vessel. The bioreactor is brought up to normal process pressure and monitored for 30 minutes in order to check for leaks. If a leak is detected, the problem is corrected and the test is run again. Once the reactor passes the test, we're ready to mix the media in the vessel.

Filtration (GFP Recovery part 5 of 5)

Although the centrifuge has removed almost all of the cell debris, some small particles still remain. We'll remove those with our final Recovery process step: Filtration. The lysate is pumped through a 0.22 (narrated as "zero-point-two-two") micron filter. This filter is fine enough that it removes virtually all of the remaining solid materials. At this point, the process stream is referred to as clarified lysate. The Recovery process is finished. The Clarified Lysate is pumped into a vented, temperature-controlled transfer vessel. This Lysate tank then moves downstream to the next series of process steps– Purification –where dissolved impurities are removed from the GFP solution and GFP is concentrated and stabilized.
